Cytokine-Driven Dendritic Maturation

1985 - 1997

The era emphasizes that maturation and generation of dendritic cells are governed by cytokine signaling, particularly granulocyte-macrophage colony-stimulating factor and inflammatory cues such as interleukin-1 and tumor necrosis factor-α, which together drive differentiation and functional maturation. Within this framework, immature dendritic cells possess robust antigen presentation capacity for native proteins, while maturation modulates processing and enhances T cell priming in vivo. The work also reveals tissue-specific dendritic cell diversity and the dynamic distribution of antigen-presenting cells in nonlymphoid tissues and draining lymph nodes, highlighting how maturation states shape immune surveillance and skin- and airway-associated responses.

Pattern: Cytokine-driven maturation and generation of dendritic cells. GM-CSF and IL-1 drive maturation of murine epidermal Langerhans cells into potent DCs; GM-CSF plus TNF-α cooperatively generate DCs; IL-1 amplifies T-dependent responses via DC function; maturation shifts DC potency and can be modulated by TNF-α; foundational work establishing cytokine-driven DC development [9], [6], [5], [1], [4].

Pattern: Antigen presentation capacity is state-dependent and demonstrates immature DCs as robust presenters of native protein; maturation alters processing; DCs pulsed with protein antigens can prime T cells in situ; soluble antigen presentation maintained by GM-CSF+IL-4 and downregulated by TNF-α; these studies map functional maturation/presentation axes [10], [17], [15], [4], [1].

Pattern: Identification, isolation, and surface phenotyping reveal DC diversity across tissues: improved isolation of human blood DCs [3]; NLDC-145 defines LC/VC/IDC groups [16]; mouse DC surface profiling by FACS across tissues [18]; tissue-localized DCs in airway/lung/pleura [13]; monocytes differentiating into CD83+ DCs [14].

Pattern: Dendritic cells drive contact-sensitivity responses and LN dynamics: skin sensitization with FITC increases LN DCs; antigen localized on DCs of draining LN; phenotypic analysis of antigen-bearing LN cells in sensitized mice [12], [2], [20].

Pattern: DCs populate nonlymphoid tissues (lung/airway) with antigen-presenting DCs; Ia+ DCs and T200 markers in pulmonary tissues; functional distribution of DCs in pulmonary tissues [7], [13].
